| Aim: This study investigated the interactive effects of interval aerobic training and mealworm extract on hepatic expression of the chronic-phase inflammatory genes MCP-1 and CCR-2, the antioxidant defense markers glutathione peroxidase and catalase, and hepatic fibrosis in rats with NAFLD. Methods: Twenty-five rats were equally allocated to five groups: 1) healthy control, 2) NAFLD, 3) exercise, 4) supplement, and 5) exercise + supplement. NAFLD was induced over 12 weeks using a diet containing high fat (15% animal fat), cholesterol (1%), and cholic acid (0.5%). The interval aerobic training protocol consisted of running for 8 weeks, 5 sessions per week, at 50% of maximal running speed for a total of 30 minutes, with weekly progressive overload. Mealworm extract was administered daily by gavage at 20 mg/kg body weight. Hepatic MCP-1 and CCR-2 gene expression was measured by Real-Time PCR, hepatic CAT and GPx activities by an enzymatic colorimetric method, and the percentage of hepatic fibrosis by Masson’s trichrome histological staining. Data were analyzed using independent-samples t-tests, two-way ANOVA, and Tukey’s post hoc test. Results: Induction of NAFLD increased hepatic MCP-1 and CCR-2 expression, reduced hepatic CAT and GPx activities, and increased the percentage of hepatic fibrosis (P≤0.001). Interval aerobic training alone significantly reversed all studied variables (P≤0.001 for all variables). Mealworm extract reduced MCP-1 expression (P≤0.001), CCR-2 expression (P≤0.001), and hepatic fibrosis (P≤0.001); however, the exercise×supplement interaction was significant only for the reduction in MCP-1 and the increase in CAT (P≤0.001 for both variables). Conclusions: Overall, interval aerobic training exerted broader effects than mealworm extract on improving NAFLD-induced inflammatory, antioxidant, and fibrotic disturbances. Nevertheless, combining exercise with the supplement may provide additional benefits by reducing MCP-1 and increasing CAT. | ||
